Semax & Selank: The Russian Nootropic Peptides — Research Overview
Semax and Selank are the two best-known nootropic peptides to come out of Russian neuroscience research. This overview is for laboratory and educational reference only.
Semax
Semax is a synthetic peptide derived from a fragment of ACTH — specifically an ACTH(4–10) analogue extended with a C-terminal Pro-Gly-Pro (PGP) tail that lengthens its effective half-life versus the unmodified fragment. In the research literature it is studied for effects on BDNF and NGF (neurotrophic factors), attention/focus, and neuroprotection.1
Mechanism (Semax)
A consistent finding across the molecular literature is that Semax rapidly increases expression of neurotrophic-factor genes. In rat glial-cell cultures, BDNF and NGF mRNA rose within roughly 30 minutes of exposure,2 and in vivo studies reported increased neurotrophin gene expression in the hippocampus and frontal cortex,3 with the time-course of NGF and BDNF expression mapped across several brain regions.4 Beyond neurotrophins, Semax is studied for modulating dopaminergic and serotonergic signaling and for neuroprotective effects in cerebral-ischemia models. These are preclinical (cell and rodent) findings.
Selank
Selank is a synthetic peptide based on the immunomodulatory peptide tuftsin. It is studied as an anxiolytic — researched for calming effects via GABA and serotonin signaling, notably without the classic sedation of some anxiolytics.5
Mechanism (Selank)
Research describes Selank's anxiolytic effect as comparable to classical benzodiazepines but without their characteristic sedation, with mechanisms studied around the GABA system; some work also implicates the opioid system in its anti-anxiety profile.6 Separately, Selank has been studied for protecting against ethanol-induced memory impairment, an effect the authors linked to changes in BDNF content in the hippocampus and prefrontal cortex.7 As with Semax, these are preclinical findings in cell and rodent models.
What the research does not establish
Both peptides carry most of their evidence from Russian preclinical and early clinical research; large, independently replicated human trials are limited, and much of the primary literature is published in Russian-language journals.
